Introduction

The Design your experiment module is divided into three core sections:

  • Cloud Software Overview: This provides the essential technical requirements and a functional guide to using the platform effectively.
  • Protein Design: This section explains how to upload protein sequences and utilise the integrated AlphaFold feature to refine your constructs on our Cloud Software
  • DNA Ordering: Here, you will find a comprehensive guide on how to use our online ordering system to secure your required DNA constructs.

Cloud Software

The Nuclera Cloud Software supports the user in the design and execution of combinatorial protein expression experiments on the eProtein Discovery platform.

  • Optimise & Validate: Access codon-optimization tools and check sequence compatibility with our eProtein Discovery System.
  • Predict & Design: Utilise the integrated AlphaFold feature to design proteins and length variants.
  • Remote Management: Design and send experiments directly to your instruments and track progress in real time.
  • Data Analysis: Review and interpret results via interactive graphs and built-in analytical tools.
  • Secure Processing: Benefit from an encrypted, cloud-based system ensuring private sequence processing.
  • Accelerate Discovery: Quickly identify the optimal conditions for solubility and expression.

This section provides a comprehensive guide to the technical requirements for ensuring your Cloud Software is fully enabled. It also includes step-by-step instructions on how to set up new projects and invite new users as an administrator.

Protein Design

You may choose to follow our Protein Variant Creation guidelines or utilise the integrated AlphaFold feature to design both proteins and their length variants.

This section provides a thorough guide on how to navigate the Cloud Software and input your specific protein of interest.

DNA order

During the experiment design phase, you will select the specific eGene™ format required for your project. Nuclera offers three distinct options to suit your laboratory's needs:

  • Linear eGene™ Fragments
  • Circular eGene™ Constructs
  • eGene™ Service

Workflows

As mentioned in the section above, when using the eProtein Discovery™ it is possible to choose between Soluble and Membrane protein workflow.

The main differences between the two workflow is how the conditions are purfied.

  • Soluble Protein Workflow: Express 192 conditions and 30 conditions are purified
  • Membrane Protein Workflow: Express and purify 88
